From “Undruggable” to Drugged: Daraxonrasib Approval Accelerates the KRAS Drug Discovery Era

The KRAS drug-discovery field has reached another major milestone.
On August 26, 2026, the U.S. FDA approved RASONQUE™ (daraxonrasib) from Revolution Medicines for adults with metastatic pancreatic adenocarcinoma who have received at least one prior systemic therapy or who are not candidates for multiagent systemic therapy. The once-daily oral drug is an inhibitor of the RAS GTPase family and represents the first approved broad RAS-targeted therapy for metastatic pancreatic cancer.
For decades, RAS—and especially KRAS—was considered one of the most difficult oncology targets to drug. The approval of daraxonrasib provides compelling validation that direct targeting of RAS-driven cancers is becoming a clinical reality.
A New Opportunity for KRAS Research
Daraxonrasib belongs to an emerging class of RAS(ON) multi-selective inhibitors designed to target active RAS signaling rather than focusing exclusively on a single KRAS mutation. Revolution Medicines reports that the approval was supported by the Phase 3 RASolute 302 study in metastatic pancreatic adenocarcinoma.
